    What is the difference between the Plus kit and the regular kit?
    A

    The Plus kit includes the functionality of detecting cross-contamination caused by positive control DNA using the Cy5 channel. This feature helps identify contamination that may occur due to positive control DNA during the experiment.

    Real-Time PCR instrument does not have the HEX dye. What should I do?
    A

    Thermofisher instruments utilize a patented dye, VIC, for the HEX wavelength range. For devices that do not support the HEX wavelength, there is a HEX plate product available that can be installed to accommodate this wavelength. Please contact the head office for product purchase and use.
    If you want to use the product without a separate purchase of the HEX plate, you can choose the VIC wavelength and experiment.

    There are two types of ROX dye in the product, which one should I use?

    The choice of ROX dye depends on the instrument you are using. Please refer to the table below to determine the appropriate ROX dye for your specific instrument.

    How should I select the channels on the PCR instrument?
    A

    For this product, which operates based on the TaqMan probe method, conduct the experiment by selecting 2 channels per well (FAM, HEX) on the PCR instrument.

    There seems to be a false positive result due to a PCR product from a previous experiment. Is there a solution? (Carry-over contamination solution)
    A

    The 2X PCR master mix includes dUTP instead of dTTP, so the UDG system is applied, and it can be solved with the following contents.

    1) Treatment of UNG (Uracil-N-glycosylase, not included in kit) in PCR products (amplicon) containing dUTP
    2) UNG hydrolyzes uracil-glycosidic bonds in the DNA regions containing dUTP, preventing re-amplification of PCR products.
    3) The UDG system ensures that the main DNA containing dTTP remains intact, preventing carry-over contamination (UDG=UNG).
